You have two options once you have set up your profile. You can be relatively passive, and wait for others to contact you first, or you can send out emails or instant messages to others whose profiles seem interesting to you. Online dating websites also offer extras like chat rooms so that you can talk to lots of people at the same time and special events, so log in often to see what's going on.
